INTENDED USE

For the qualitative determination of anti-insulin (IAA), islet cells (ICA), glutamate decarboxylase (GAD), tyrosine phosphatase (IA-2) and zinc transporter 8 (ZnT8) in human serum. antibody.

The above antibodies are highly correlated with type 1 diabetes. Combined detection has important clinical significance for the diagnosis, prediction and improvement of the diagnosis of diabetes. Clinically used to assist in the diagnosis of autoimmune diabetes.

Autoantibodies have emerged early in the natural course of autoimmune diseases and are characterized by little or no symptoms for a long time in the preclinical phase. Moreover, autoantibodies that accompany the clinical progression of autoimmune diseases often persist throughout the course of the disease. Most of the disease is initially resistant to one or a small amount of autoantibodies, which can then develop into autoantibodies to many other autoantigens or their antigenic sites. Autoantibody serological testing is thought to have a predictive effect on autoimmune diseases because autoantibodies reflect the appearance, nature, and strength of autoimmune responses. The prospect of multiple autoantibody detection systems (multiple joint tests) for preventive screening of open and high-risk populations has important clinical implications.

PRINCIPLE

The test is based on the principle of the line immune assay (LIA).

IAA, ICA, GAD, IA2 and ZnT8 autoantigens are applied as lines on a nitrocellulose membrane. The nitrocellulose membrane is blocked to prevent unspecific reactions. During incubation of a strip with diluted patient samples, autoantibodies present in the sample will bind to the antigens on the strip. For the detection of the bound antibodies, an alkaline phosphatase labelled antihuman IgG antibody is used. After addition of the substrate solution, the appearance of purple blue lines indicates the existence of (auto) antibodies against the respective antigens.
